Using molecular network analysis to understand current HIV-1 transmission characteristics in an inland area of Yunnan, China

HIV-1 molecular surveillance provides a new approach to explore transmission risks and targeted interventions. From January to June 2021, 663 newly reported HIV-1 cases were recruited in Zhaotong City, Yunnan Province, China. The distribution characteristics of HIV-1 subtypes and HIV-1 molecular network were analysed. Of 542 successfully subtyped samples, 12 HIV-1 strains were identified. The main strains were CRF08_BC (47.0%, 255/542), CRF01_AE (17.0%, 92/542), CRF07_BC (17.0%, 92/542), URFs (8.7%, 47/542), and CRF85_BC (6.5%, 35/542). CRF08_BC was commonly detected among Zhaotong natives, illiterates, and non-farmers and was mostly detected in Zhaoyang County. CRF01_AE was frequently detected among married and homosexual individuals and mostly detected in Weixin and Zhenxiong counties. Among the 516 pol sequences, 187 (36.2%) were clustered. Zhaotong natives, individuals aged ≥60 years, and illiterate individuals were more likely to be found in the network. Assortativity analysis showed that individuals were more likely to be genetically associated when stratified by age, education level, occupation, and reporting area. The genetic diversity of HIV-1 reflects the complexity of local HIV epidemics. Molecular network analyses revealed the subpopulations to focus on and the characteristics of the risk networks. The results will help optimise local prevention and control strategies.
Disrupted dynamic functional connectivity of hippocampal subregions mediated the slowed information processing speed in late-life depression

Background
Slowed information processing speed (IPS) is the core contributor to cognitive impairment in patients with late-life depression (LLD). The hippocampus is an important link between depression and dementia, and it may be involved in IPS slowing in LLD. However, the relationship between a slowed IPS and the dynamic activity and connectivity of hippocampal subregions in patients with LLD remains unclear.
MethodsOne hundred thirty-four patients with LLD and 89 healthy controls were recruited. Sliding-window analysis was used to assess whole-brain dynamic functional connectivity (dFC), dynamic fractional amplitude of low-frequency fluctuations (dfALFF) and dynamic regional homogeneity (dReHo) for each hippocampal subregion seed.
ResultsCognitive impairment (global cognition, verbal memory, language, visual–spatial skill, executive function and working memory) in patients with LLD was mediated by their slowed IPS. Compared with the controls, patients with LLD exhibited decreased dFC between various hippocampal subregions and the frontal cortex and decreased dReho in the left rostral hippocampus. Additionally, most of the dFCs were negatively associated with the severity of depressive symptoms and were positively associated with various domains of cognitive function. Moreover, the dFC between the left rostral hippocampus and middle frontal gyrus exhibited a partial mediation effect on the relationships between the scores of depressive symptoms and IPS.
ConclusionsPatients with LLD exhibited decreased dFC between the hippocampus and frontal cortex, and the decreased dFC between the left rostral hippocampus and right middle frontal gyrus was involved in the underlying neural substrate of the slowed IPS.

Korean pine is an economically essential afforestation species limited by the unreasonable collection of cones, indiscriminate use of chemical pesticides and pest damage. This study aimed to determine whether spraying bacterial or fungal solutions affected insect pests, cone development, and the seed quality of Korean pine Pinus koraiensis Sieb. et Zucc. The experiment was conducted in a forest plantation in Linkou County (Heilongjiang, China) in 2019. Four fungal strains and one bacterial strain were applied during the flowering phase of Korean pine. The results after a year and a half of study indicated that a high concentration of Bacillus thuringiensis 223176 promoted cone development, increased seed weight, and reduced the proportion of damaged cones. Under this treatment, there were 15.873% damaged cones; the seed weight reached 0.829 g, and there were 82.738% fully developed cones. Trees treated with the second most effective strain, Beauveria bassiana 122077, had 30.556% damaged cones and an average seed weight of 0.810 g. Leucanicillium antillanum 01 performed the worst in this study. The seed weight was only 0.775 g, and the damaged and fully developed cones were 52.444 and 41.773%, respectively. In summary, spraying bacterial or fungal solutions during the flowering stage of Korean pine positively impacted seed quality and effectively decreased damage by the lepidopteran species that feed on the cones and seeds in this study.
AGPAT3 Gene polymorphisms are associated with milk production traits in Chinese Holstein cows

The current study reports the identification of previously undiscovered single-nucleotide polymorphisms (SNPs) in the bovine AGPAT3 gene and further investigates their associations with milk production traits. Our results demonstrate that the major allele C of the SNP g.12264 C > T is positively correlated with test-day milk yield, protein percentage and 305-day milk yield. Importantly, in silico analysis showed that the C/T transition at this locus gives rise to two new transcription factor binding sites (TFBS), E2F1 and Nkx3-2. Polymorphism g.18658 G > A was the only SNP associated with milk urea nitrogen (MUN) with the G allele related to an increase in milk urea nitrogen as well as fat percentage. The GG genotype of SNP g.28731 A > G was associated with the highest fat and protein percentage and lowest 305-day milk yield and somatic cell score (SCS). The association between AGPAT3 locus and milk production traits could be utilized in marker-assisted selection for the genetic improvement of milk production traits and, probably in conjunction with other traits, for selection to improve fitness of dairy cattle.
Prediction of parental alienation on depression in left-behind children: A 12-month follow-up investigation

Aims
Alienation towards parents often occurs when parents divorce; however, it can also occur when one or both parents leave for work for more than 6 months. Our previous investigation has confirmed a high level of feelings of alienation towards parents among Chinese left-behind children. However, the longitudinal prediction of alienation on children's mental health outcomes remains largely unknown. This study aims to observe the prediction of alienation towards parents on children's depression 12 months later and potential mediators and moderators.
MethodsA total of 1090 Chinese left-behind children took part in this 12-month follow-up investigation, using the Chinese version of the Inventory of Alienation towards Parents (IAP), the Children's Depression Inventory (CDI), the Adolescent Self-Rating Life-events Checklist (ASLEC), and the Adolescent Resilience Scale.
ResultsAlienation towards parents was high (16.42 ± 7.27 for mother, 15.63 ± 7.17 for father) in left-behind children, and 21.01% of children reported depression. Alienation towards parents predicted current depression of children directly and later depression indirectly; children's alienation toward their mothers was a stronger predictor of depression than alienation towards fathers. In models, stressful life-events acted as a risk mediator. Previous depression was the strongest risk predictor, resilience was the strongest protective factor, and duration of fathers’ absence and parents’ marital status moderated the predictive effects.
ConclusionsThis study is among the first to longitudinally confirm that alienation towards parents is a predictor of children's later depression. The results provide important suggestions for families and schools; i.e. to prevent depression in left-behind children, parent−child bonds especially alienation towards mothers, should be carefully considered, and individuals with more negative life-events and weaker resilience need further attention.
The characteristics of pretreatment HIV-1 drug resistance in western Yunnan, China

HIV-1 drug resistance can compromise the effectiveness of antiretroviral therapy (ART). A survey of pretreatment HIV-1 drug resistance (PDR) was conducted in Lincang Prefecture of Yunnan Province. From 372 people living with HIV/AIDS initiating ART for the first time during 2017–2018, 322 pol sequences were obtained, of which 11 HIV-1 strain types were detected. CRF08_BC (70.2%, 226/322) was the predominant strain, followed by URF strains (10.6%, 34/322). Drug resistance mutations (DRMs) were detected among 34.2% (110/322) of the participants. E138A/G/K/R (14.3%, 46/322) and V179E/D/T (13.7%, 47/322) were the predominant DRMs. Specifically, E138 mutations commonly occurred in CRF08_BC (19.9%, 45/226). Among the DRMs detected, some independently conferred resistance, such as K65R (1.6%, 5/322), Y188C/F/L (0.9%, 3/322), K103N (0.6%, 2/322) and G190A (0.3%, 1/322), which conferred high-level resistance. The prevalence of PDR was 7.5% (95% CI: 4.6–10.3%) and the prevalence of non-nucleotide reverse transcriptase inhibitor (NNRTI) resistance was 5.0% (95% CI: 2.6–7.4%), which is below the threshold (⩾10%) of initiating a public health response. In conclusion, HIV-1 genetic diversity and an overall moderate level of PDR prevalence were found in western Yunnan. PDR surveillance should be continually performed to decide whether a public health response to NNRTI resistance should be initiated.

Thermal perception is crucial for the fitness of marine invertebrates in intertidal and shallow waters. TRPA1 is a non-selective cation channel that belongs to the TRP family with pivotal roles in initiating signal transduction of thermal perception. We investigated expression patterns of SiTRPA1 in different tissues (tube feet, coelomocytes, gonads and gut) of the sea urchin Strongylocentrotus intermedius. SiTRPA1 expression patterns under acute and long-term temperature stimuli were investigated in tube feet of sea urchins. In the present study, the highest expression of SiTRPA1 was detected in tube feet of S. intermedius. The SiTRPA1 expression level in tube feet were significantly 235.7-fold, 450.0-fold and 3299.7-fold higher than those in the coelomocytes, gonads and gut (df = 3, F = 47.382, P < 0.001). Expression levels of SiTRPA1 in the other tissues (coelomocytes, gonads and gut) were not significantly different (df = 3, F = 47.382, P = 0.972). There was no significant difference of SiTRPA1 expression among all groups in the acute temperature increase experiment (df = 4, F = 0.25, P = 0.902). In the acute temperature decrease experiment, the expression of SiTRPA1 showed no significant difference among all groups (df = 4, F = 1.802, P = 0.205). With long-term exposure (6 weeks) to different temperatures, SiTRPA1 expression in the low temperature group (10°C) was significantly higher than those in the high temperature (20°C) and the control groups (15°C) (df = 2, F = 9.57, P = 0.014). There was no significant difference of SiTRPA1 expression between the high temperature (20°C) and the control temperature (15°C) groups (df = 2, F = 9.57, P = 0.808). These results indicate that SiTRPA1 expression significantly responds to long-term low temperature but not to acute temperature decrease. The present study provides new insights on the distribution and temporal expression of TRPA1 in marine invertebrates after acute and long-term temperature stimuli.
Implications of radiocarbon ages of organic and inorganic carbon in coastal lakes in Florida for establishing a reliable chronology for sediment-based paleoclimate reconstruction

Coastal lake sediments are valuable paleoclimate archives provided that they can be accurately dated. Here, we report radiocarbon ages of bulk sediment organic matter (OM), plants, shells, particulate OM, and dissolved OM from coastal lakes in Florida. Bulk sediment OM yielded ages that are consistently older than contemporaneous plants and shells, indicating significant radiocarbon deficiencies in sedimentary OM in these lakes. The data show that the OM radiocarbon deficiency varies over time and with location, making it impossible to determine a proper correction factor for radiocarbon ages of bulk sediments from these lakes. As a result, we consider ages obtained from bulk sediment OM from these lakes unreliable. The age reversals in bulk sediment OM observed in the sediment cores are likely caused by rapid increases in erosion and sedimentation resulting from large storm events. The data also show that sedimentation rate can vary considerably within a given lake, implying that an age-depth model established for one core cannot be directly applied to other cores despite their close proximity. Analyses of shells from one of the lakes suggest that fresh/brackish-water shells may serve as a good substrate for radiocarbon dating owing to a small reservoir effect on inorganic carbon.
